    J'ai fait des ?tudes de Sciences Naturelles suivies d?une th?se de 3e cycle puis d?un doctorat d??tat en Embryologie, soutenu en 1983 ? Strasbourg chez le professeur Jean Thiebold. Au d?but des ann?es 80, l'Embryologie ?tait une discipline ? bout de souffle, probablement ? cause du besoin de techniques nouvelles comme la biologie mol?culaire ou la g?n?tique. J?ai alors effectu? plusieurs stages pour apprendre ces techniques et ? mon retour, j?ai cr?? avec Dani?le Hoffmann un petit groupe de recherche d?di? ? l?Immunit? dans le Laboratoire d?Endocrinologie des Insectes dirig? par Jules Hoffmann.
    Nous avons alors choisi la mouche noire Phormia terranovaecomme mod?le d'?tude car la larve et l'adulte de cette mouche vivent sur la viande en d?composition, ce qui ? priori laisse supposer des m?canismes de d?fense anti-microbiens tr?s efficaces. De plus cette mouche est phylog?n?tiquement proche de la Drosophile que nous envisagions d?j? comme mod?le ? cause de sa g?n?tique. En 1985, nous avons entam? nos recherches sur le syst?me immunitaire de la drosophile et au cours de la p?riode qui a suivi, plusieurs moments charni?res sont ? noter :
   
    Au cours des ann?es 1987 et 1988, j'ai pass? 18 mois dans ce qui ?tait l'une des premi?res compagnies de biotechnologie fran?aise : Transg?ne SA. Pendant cette p?riode, j'ai r?ussi ? exprimer dans des syst?mes h?t?rologues, les peptides antimicrobiens  isol?s dans la mouche. L'exp?rience acquise dans cette compagnie a ?t? extr?mement pr?cieuse  lorsque j'ai particip?, en 1999, ? la cr?ation de l?entreprise Entomed SA.
    En 1991, mon petit groupe avait obtenu des r?sultats tr?s encourageants et progressivement, tout le laboratoire, environ 40 personnes, allait r?orienter leur projet de recherche de l?endocrinologie ? l?immunit? des Insectes. En 1992, gr?ce ? mon pass? en embryologie, j?ai ?t? nomm? professeur de Biologie du D?veloppement, et par go?t, autant que par devoir, j'ai commenc? ? m'int?resser aux probl?mes du d?veloppement pr?coce et ? la formation des axes chez la drosophile. Ceci m'a amen? en 1996 ? la d?couverte de l'implication d?une voie de transduction de signal, appel? voie Toll, (connue ? l'origine pour son intervention dans la mise en place de l'axe dorso-ventral au cours de l?embryogen?se pr?coce) dans le syst?me immunitaire de la drosophile. Cette d?couverte (Lemaitre et al. Cell, 1996) a permis de trouver les homologues humains de Toll, les Toll-like R?cepteurs (TLRs) , et a modifi? profondement notre vision du Syst?me Immunitaire Inn?.
    Les TLRs humains appartiennent ? une famille de r?cepteurs qui reconnaissent directement les d?terminants microbiens et activent le syst?me immunitaire. Au cours du d?veloppement pr?coce, Toll de Drosophile reconna?t un ligand qui doit ?tre activ? par des prot?ases ? s?rine qui ne sont pas n?cessaires pour l?activation du syst?me immunitaire chez la mouche adulte. On pouvait donc se demander comment ce r?cepteur ?tait activ? au cours de la r?ponse immunitaire. En 1998-1999, au cours d?une ann?e sabbatique dans le D?partement de G?n?tique ? l?Universit? de Cambridge (Royaume Uni), j?ai d?couvert qu?une mutation perte de fonction (appel?e ? necrotic ?) dans un inhibiteur de prot?ase ? serine, une serpine, montrait une activation constitutive du syst?me immunitaire de la mouche (Levashina et al., Science, 1999). Cette d?couverte nous a permis de comprendre que chez la drosophile, la reconnaissance des microbes n?cessite des r?cepteurs situ?s en amont de Toll dans le syst?me circulatoire ouvert des insectes. Nous avons d?couvert ces r?cepteurs en 2001 et les avons identifi?s comme des mol?cules reconnaissant le peptidoglycane des parois bact?riennes (Michel et al., Nature, 2001). Ces ?v?nements de reconnaissance sont ensuite transmis vers Toll par l?interm?diaire de cascades de prot?ases ? s?rine, analogue des cascades de la coagulation sanguine ou du compl?ment. Nous avons commenc? ? isoler certaines de ces prot?ases (Ligoxygakis et al., Science, 2002). R?cemment et en se basant sur une de mes observations anciennes, nous avons pu montrer que l?une de ces prot?ases agissait comme un senseur direct pour des facteurs de virulence prot?olytiques excr?t?s par des champignons pathog?nes (Gottar et al., Cell, 2006; El Chamy et al., Nature Immunology, 2008).
