B cell tolerance and autoimmunity

Although our understanding of the mechanisms of immunological tolerance, especially in mice, have improved greatly over recent years, the clinical translation to the prevention and cure of autoimmune diseases is so-far disappointing. Human autoimmune diseases are thought to arise from various combinations of genetic, environmental and stochastic (for example, immune repertoire generation) factors. In our group we study the two first factors using different approaches :

  • The identification of B cell genetic defects in patients with systemic lupus erythematosus

  • The study of B cell tolerance breakdown, by an analysis of autoreactive B cell phenotype and response to infectious agents
